电针对亚急性衰老大鼠肝与血清SOD MDA T-AOC的影响 被引量:14

摘要 目的:观察电针对D-半乳糖造成的亚急性衰老大鼠肝与血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、总抗氧化能力(T-AOC)的影响,并光镜下观察肝、胸腺组织的变化,从自由基学说、组织学探讨电针延缓衰老的机制。方法:将48只Wistar大鼠随机分为四组,其中模型组、电针组、维生素E组用D-半乳糖制备衰老模型,空白组则注射等量生理盐水。造模同时进行治疗,电针组选用"百会"、"关元"、"足三里"、"三阴交",每日刺激10min,维生素E组造模同时灌服维生素E植物油溶液。42天后,采集标本,按试剂盒要求测试各项指标,病理切片HE染色观察组织病理学变化。结果:与空白组相比,模型组SOD活性、T-AOC明显降低,MDA含量明显升高;与模型组相比,电针组、维生素E组均能显著提高SOD活性、T-AOC,降低MDA含量;电针组肝组织SOD活性与维生素E组有显著性差异。病理切片提示电针组、维生素E组肝及胸腺较空白组仅轻微改变,少量脂肪变性,明显好于模型组。结论:电针可以通过抗自由基延缓衰老,并可改善肝组织及胸腺组织脂肪变性。 Objective: To explore the effects of electricalLacup^uncture on SOD,MDA,T-AOC in serum and liver tissue due to acute senile rat model, and the structural change of liver and thymus tissue was observed. To research the anti-aging mechanism of electrical-acupuncture from free radical theory and histology. Method: 48 Wistar rats divided into four groups randomly:Model group, Electrical-Acupuncture group, VitE group were injected D-gal to build acute senile rat model, Blank group was injected with saline of the same volume. Treatment was given meanwhile model establishment. Electrical-Acupuncture group was pricked : baihui ( GV 20 ) and guanyuan ( CV 4 ), zusanli ( ST 36 ) two sides, sanyinjiao (SP 6) two sides for three rotation, once daily,10min. VitE group received VitE oil daily. All rats were sacrificed after 6 weeks, observe and analyze SOD,MDA,T-AOC with serum and liver tissue. Pathological sections were dyed to observe the histopathological changes. Result : Compared with blank group, the activity of SOD and T-AOC decreased significantlv, the level of MDA increased sienificantlv in model groud. Compared with model group, the activity of SOl) and T-AOC increased significantly, and the level of-Mr)'A decreased significantly in Electrical-Acupuncture group and VitE group. Compared with VitE group, the activity of SOD in liver tissue increased significantly in Electrical-Acupuncture group. Pathological section suggested that, compared with blank group, there was little changes with fatty degeneration of liver and thymus in electrical-acupuncture group and VitE group. It was better than model group obviously. Conclusion: electrical- acupuncture could improve the D-gal induced decay process by anti free radical, and improve the liver and thymus tissue from fatty degeneration.
