摘要
目的探讨胃癌组织胸苷酸合成酶(TS)表达水平与5-氟尿嘧啶(5-Fu)耐药性的关系。方法用ATP-TCA法对35例胃癌患者新鲜标本进行5-Fu体外药敏检测;免疫组化法检测其TS表达水平,比较分析不同TS表达水平与体外药敏有效率的关系。结果TS高表达组有效率18.8%(3/16);TS低表达组有效率52.9%(9/17)。TS高、低表达组对5-Fu敏感性差异有统计学意义(P<0.05)。结论胃癌组织TS表达水平与5-Fu敏感性可能有关,TS的表达水平检测可能有助于预测胃癌组织对5-Fu的敏感性。
Objective To investigate correlation between the level of thymidylate synthase and the antitumor activities of 5-fluorouracil (5-Fu) determined by ATP sensitivity test in vitro. Methods The gastric carcinoma cells taken from fresh samples of 35 patients with the cancer were cultured in vitro to test the sensitivity of these cells to 5-Fu with the method of ATP-TCA. The level of TS was detected by means of immunohistochemistry,and the correlation between the level of TS and chemosensitivity assay was then analyzed. Results The response rate of the highlevel TS group to 5-Fu was 18.8%(3/16) and was significantly lower than that of the low-level TS group(52.90%,9/17,P〈0.05).Conclusion The results in the present study suggest that the level of TS is associated with the antitumor activities of 5-fluorouracil, indicating that the level of TS might be used as stratification maker for predicting the antitumor activities of 5-fluorouracil.
