The effect of interferon α with different structures on ACHN tumor suppression

摘要 目的比较国产长效干扰素PEG IFN α-1b与原型IFNα-1b及进口PEG IFN α-2b对荷瘤裸鼠的治疗效果。方法以对数生长期的人肾腺癌ACHN细胞接种于雄性BALB/c裸鼠体内制备荷瘤小鼠模型,接种后4周按肿瘤大小将ACHN荷瘤鼠随机分为溶剂对照、PEG IFN α-1b、PEG IFN α-2b和IFN α-1b4个组。溶剂或受试药均连续5周皮下注射给药,给药后每周1次测量动物体重及肿瘤体积。给药后第5周末将动物全身麻醉后处死,称量肿瘤重量,并计算肿瘤抑制率。结果显示第5周末PEG IFN α-1b、PEG IFN α-2b和IFN α-1b组的肿瘤抑制率分别为37.68%、44.31%、48.10%,与溶剂对照组相比,差异均有统计学意义(P<0.05)。结论3种α干扰素对裸鼠体内ACHN肿瘤生长均有明显地抑制作用。 Objective To evaluate the anti-tumor activity of interferon a with different structures including pegylated interferon α-lb (PEG IFN α-lb), pegylated interferon α-2b (PEG WN α-2b) and natural interferon α (IFN-α) in male BALB/C nude mice with human renal adenearcinoma (ACHN) tumor. Methods Athymic BALB/C nude mice were received a subcutaneous injection of ACHN cells (2 × 10^6/0. 2ml) on the rear flantc After four weeks, the size of tumor reached a mean volume of 50-100 mm^3, and the interferon was administrated. The mice with tumor were divided into four groups, and were administrated with placebo (0. 2ml/mouse thrice weekly), IFN a-lb (50μg/mouse thrice weekly), PEG IFN α-lb (150μg/mouse once weekly) and PEG IFNα-2b (150μg/mouse once every week), respectively, for five weeks. The tumor volumes were measured weekly prior to treatments until the end of the fifth week. All mice in each group were sacrificed to measure the weight of the residual tumor. The anti-tumor activity of IFNα in vivo was evaluated by studying their ability to reduce the volume and weight of tumor. Results IFNα may significantly reduce the volume and weight of ACHN tumor in mice. The average tumor weight was 0. 422g in placebo group, while 0. 263g, 0. 235g and 0. 219g in the groups treated with PEG IFN α-lb, IFN α- lb and PEG IFN α-2b, respectively. No significant difference was found among the three groups treated with IFNα. Moreover, the volume of tumor in three groups treated with IFNα was significant reduced than that in placebo group. Conclusion IFNα with different structures have showed the similar ability of ACHN tumor suppression at the dosage of 150μg per week.
