白藜芦醇对氧化型LDL所致血管内皮细胞损伤的保护作用 被引量:19

摘要 目的探讨白藜芦醇对血管内皮细胞脂质过氧化损伤的保护作用及其可能的分子机制。方法在铜离子诱发低密度脂蛋白(LDL)氧化修饰的基础上,建立内皮细胞脂质过氧化损伤模型,将细胞分为7组,即正常对照组,溶剂(DMSO)对照组,oxLDL氧化损伤组,oxLDL加槲皮素阳性对照组,oxLDL加白藜芦醇低剂量(0.5μmol/L)、中剂量(5μmol/L)、高剂量(50μmol/L)组,其中后4组将200μg/ml的oxLDL作用于以槲皮素及不同浓度白藜芦醇预培养24h的内皮细胞,然后继续培养18h。检测各组内皮细胞培养上清液和细胞匀浆中丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,并测定细胞凋亡率。结果oxLDL作用后血管内皮细胞损伤明显,细胞培养上清液和细胞匀浆中MDA含量升高,SOD、GSH-Px活性下降,凋亡率可达41.4%±2.3%,而加入白藜芦醇预培养可明显减轻oxLDL对抗氧化酶SOD、GSH-Px的影响,降低MDA含量,并显著减少凋亡细胞数量,ox-LDL加白藜芦醇低剂量组内皮细胞凋亡率降至13.0%±1.5%,各指标差异有显著性(P<0.01)。结论白藜芦醇可保护和修复ox-LDL引起的血管内皮细胞损伤,其作用机制可能与抗氧化作用有关。 Objective To study the protective effect of resveratrol on endothelial cells injury induced by oxidized low-density lipoprotein (oxLDL) and its protective mechanism Methods The injured model was established by endothelial cell treated with oxLDL. In the experiment, endothelial cell were cultured in vitro and divided into seven groups which were: control group; solvent (DMSO) group, ox- LDL group, quercetin plus oxLDL group, Reseveratrol-L (0. 5 mol/L), M(5 mol/L), H(50 mol/L) plus oxLDL group respectively. Endothelial cells were incubated with 200μg/ml oxLDL for 18 hours in the absence or presence of various concentrations of resveratrol and quercetin. The apoptotic index was detected by flow cytometric. Meanwhile, the content of Malondialdehyde (MDA), and the activity of SOD, GSHPX in the culture medium and homogenate of endothelial cells were measured. Results Incubation of endothelial cells with ox- LDL for 18 h elicited a significant increase in the percent of apoptotic endothelial cells, in control group, the percent of apoptotic ceils is 7. 6 ±0. 6, however in oxLDL group, it increased to 41.4±2. 3, furthermore, oxLDL induced the increase of the contents of MDA, and accompanied by the decrease in the activity of SOD, GSHPX in the endothelial cells, the difference of was significant as compared with control group. Pretreatment of resveratrol with 0. 5 ~50 mol/L decreased these oxLDL-induced changes, in Reseveratrol with 50 mol/L group, the percent of apoptotic cells decreased to 13. 0 ± 1.5, the difference of was significant as compared with oxLDL group (P〈0. 01). Conclusion Resveratrol posses a protective effect against endothelial cells injuries induced by oxLDL, which is probably via anti-oxidation, inhibiting the apoptosis of endothelial cells.
