摘要
目的:测定NK细胞表面激活性受体NKG2D、NKp46和抑制性受体NKG2A、杀伤细胞免疫球蛋白样受体(KIR)在尖锐湿疣中的表达。方法:采用四色免疫荧光抗体染色,经流式细胞仪检测30例尖锐湿疣患者外周血NK细胞表达NKG2D、NKp46、NKG2A和KIR的水平,并以40例正常人作为对照。结果:尖锐湿疣患者外周血NK细胞NKG2D、NKp46分子表达水平显著低于对照组(P<0.001及P<0.01),而抑制受体NKG2A、KIR的表达水平差异无统计学意义(P>0.05)。结论:人乳头瘤病毒感染能下调NK细胞激活性受体表达、抑制NK细胞的活化,使病毒感染细胞逃逸其天然杀伤作用。因此,NK细胞的激活性和抑制性受体表达失衡在尖锐湿疣的发病机制中起着重要的作用。
Objective: To investigate the expression of activating (NKG2D, NKp46) and inhibitory (NKG2A, KIR) natural killer cells receptors in peripheral blood NK cells of patients with condyloma acuminatum. Methods: The expression of NKG2D, NKG2A, KIR and NKp46 NK cell receptors were measured by flow cytometry with four - color immunofluorescent staining in 30 patients with condyloma acuminatum and 40 normal controls. Results: The expression of NKG2D and NKp46 NK cells was significantly decreased in condyloma acuminatum patients compared with that in normal controls ( P 〈 0.001 and P 〈 0.01, respectively). No statistically significant differences were found in the expression of NKG2A and KIR receptors between the patients and controls ( P 〉 0.05). Conclusions: The expression of activating receptors on NK cells can be down- regulated by HPV infection, resulting in the hampering NK cell activation and escaping from natural cytotoxic against viral- infected cells. The altered expression of NK receptors is likely to result in the imbalance of activating and inhibitory receptors that may play an important role in the regulation of NK cell activities and pathogenesis of condyloma acuminatum.
