摘要 由特异蛋白激酶和蛋白磷酸酶共同参与的蛋白质可逆性磷酸化是细胞生物学过程的重要调节机制,蛋白磷酸酶2A是真核生物主要的丝氨酸/苏氨酸蛋白磷酸酶,具有复杂的亚基构成,参与细胞的各种调节机制,包括信号转导途径、细胞周期进程、DNA复制、基因转录和蛋白质翻译等,其功能受多种层次的调控。最新的研究揭示了PP2A的三维晶体结构以及甲基化在全酶组合中的作用,同时对PP2A在细胞转化方面的功能与机制有了更深入的阐释。 Reversible protein phosphorylation by specific kinases and phosphatases is an important regulatory mechanism in many cellular processes. Protein phosphatase 2A (PP2A), a major serine/threonine phosphatase in eukaryotic cells, has complex subunit composition and is involved in cellular regulatory mechanisms, including signal transduction, cell cycle progression, DNA replication, gene transcription and protein translation. It is regulated at different levels. This article reviews the latest studies on the three-dimensional crystal structure of PP2A, the role of methylation in the formation of holoenzymes and cell transformation.
