摘要
目的探讨系统性红斑狼疮(SLE)患者外周血B淋巴细胞CD40和肿瘤坏死因子受体相关因子1(TRAF1)的表达及两者间的相互关系。方法采用免疫荧光标记和流式细胞仪技术检测28例SLE患者及14名正常人外周血B细胞CD40、TRAF1的表达。结果(1)活动期SLE患者外周血B细胞CD40、TRAF1的表达高于缓解期SLE患者及正常对照组,差异有高度统计学意义(P<0.01)。缓解期SLE患者外周血B细胞CD40的表达与正常对照组比较,差异无统计学意义(P>0.05);但TRAF1的表达高于正常对照组,差异有统计学意义(P<0.05)。(2)活动期和缓解期SLE患者外周血B细胞CD40的表达与TRAF1的表达成正相关(P<0.05)。结论外周血B细胞CD40、TRAF1在SLE的发病机制中可能参与免疫调节作用。
Objective To investigate whether the expression of CD40 and TNF receptor associated factorl(TRAF1) in peripheral B lymphocytes of systemic lupus erythematosus(SLE) patients is abnormal and also its relationship. Methods The expression of CD40 and TRAF1 in peripheral B lymphocytes in 28 patients with SLE and in 14 normal controls was examined by immunofluorescence and flowcytometry. Results (1) The ratio between positive CD40 and TRAF1 in peripheral B lymphocytes was higher in patients with active SLE than that in patients with inactive SLE and normal controls (P〈0.01), and there was no difference in the ratio of CD40 between patients with inactive SLE and the normal controls (P〉0.05).The rate between positive TRAF1 in peripheral B lymphocytes is higher in patients with inactive SLE than that in the normal controls (P〈0.05). (2) In patients with active SLE and inactive SLE, there was positive correlation between the rate of positive CD40 in peripheral B lymphocytes and the rate of positive TRAF1. Conclusion CD40 and TRAF1 in peripheral B lymphocytes may play an immunoregulator role in the pathogenesis of SLE.
