血清药理学与膜片钳技术对川芎扩冠离子机制的探讨 被引量:1

摘要 目的通过观察分析川芎灌胃动物后分离的含药血清(含川芎血清)对猪冠状动脉血管条及平滑肌细胞BKCa的作用,旨在从细胞分子水平阐述其扩冠机制,也为探索建立血清药理学与膜片钳技术结合于中药研究的联合技术奠定基础。方法用膜片钳实验及冠状动脉血管条实验观察含川芎血清、川芎主要活性成分川芎嗪(阳性对照)和空白血清(阴性对照)对BKCa的作用以及对离体猪冠状动脉血管条的影响。结果在内面向外式和贴附式膜片上,含川芎血清和空白血清对BKCa通道无激活作用,但在内面向外式膜片上,阳性对照川芎嗪对BKCa通道具有激活作用,且川芎嗪具有对抗高钾(45mM)去极化所致的冠脉血管条收缩的作用,含川芎血清和空白血清无此作用。结论川芎嗪对BKCa通道具有直接激活作用,含川芎血清在体外实验中对BKCa通道无明显激活作用。 Objective By studying the effects of the extracted serum after administering orally chuanxiong to mice or rats( serum containing chuanxiong) on porcine coronary artery strips and BKCa channels of coronary smooth muscle cells, the study was designed to expound the mechanisms of chuanxiong in dilating coronary artery on cellular and molecular level and to do the groundwork for the combination of serum pharmacology with electrophysiology method. Methods After preparing the serum containing chuanxiong, We observed the effects of serum containing chuanxiong on large conductance Ca^2+ -activated K^+ Channel( BKCa ) and strips of coronary smooth muscle. Results In cell-attached and inside-out patches, Serum containing chuanxiong and Serum without chuanxiong had no effects on BKCa channels in vitro, in inside-out pathces, BKCa channels could be activated by the positive control TMP, and TMP could attenuate high K^+ - evoked contraction of porcine coronary smooth muscle strips. Both serum without chuanxiong and serum containing chuanxiong had no influence on coronary artery tone. Conclusion TMP can directly activate BKCa channels in porcine coronary artery smooth muscle cells. Serum containing chuanxiong had no effects on BKCa channels in vitro.
