摘要
目的:探讨天子饮对偏头痛大鼠脑干组织G蛋白亚基(Gsα和Giα)含量的影响。方法:40只SD大鼠随机分为4组各10只。正常组大鼠不作任何处理,盐水组大鼠皮下注射盐水2 mg/Kg;模型组大鼠皮下注射硝酸甘油(10 mg/Kg)建立实验性偏头痛大鼠模型;天子饮组大鼠以天子饮灌胃连续21 d后造模建立偏头痛模型。第22天时4组大鼠均处死,运用免疫印迹法(Western blot)检测脑干组织刺激性Giα和抑制性Gsα的含量。结果:模型组和天子饮组大鼠皮下注射硝酸甘油4 h后脑干组织Gsα含量与对照组及盐水组比较明显升高,Giα蛋白含量明显降低(均P<0.01),Gsα/Giα蛋白比值升高;与模型组比较,天子饮组脑干组织中Gsα蛋白含量明显降低,Giα蛋白含量明显升高(均P<0.01),Gsα/Giα蛋白比值降低。结论:偏头痛发作可能与大鼠脑干组织G蛋白信号传导系统功能障碍有关,天子饮介导的G蛋白信号通路可能是其防治偏头痛的重要机制之一。
Objective: To explore the effect of Tianzi decoction on the content of G protein in the brain stem of migraine rats. Methods: The rat migraine model was established by subcutaneous injection of glycerol trinitrate. Forty rats were randomly divided into 4 groups: normal control group, normal saline group,model control group and Tianzi decoction group. The content of Gia and Gsα protein in the rat brain stem was detected by using Western blot method. Results: The content of Gsa protein in the brain stem of the model control group was significantly increased (P〈0.01) ,while the Gia protein was significantly reduced (P〈 0.01) ,and the ratio of Gsα/Giα was increased as compared with normal control group. The Gsa protein in the rat brain stem of the Tianzi decoction group was significantly reduced as compared with model control group (P〈0.01) ,and Gia protein level was slightly higher in Tianzi decoction group than in model control group (P〈0.01), resulting in the decreased Gsα/Giα ratio. Conclusion: The results provided important information for understanding how G-protein-linked signaling system played an important role in the migraine attach. Tianzi decoction-mediated G-protein-linked signal transduction pathway may be one of the important mechanisms by which Tianzi decoction prevent and treat migraine.
