星洲银罗非鱼线粒体DNA的RFLP研究 被引量:2

摘要 用EcoRⅤ、MboⅡ、NaeⅠ、PvuⅡ、StyⅠ、KpnⅠ、SacⅠ、HindⅢ、DraⅠ、HpaⅡ等10种限制性核酸内切酶对星洲银罗非鱼线粒体DNA(mtDNA)进行酶切分析,其中StyⅠ、SacⅠ和DraⅠ表现出多态性,HpaⅡ酶切后产生零碎片段,EcoRⅤ、NaeⅠ、PvuⅡ、KpnⅠ只有一个切点,HindⅢ有两个切点,MboⅡ有5个切点但有小片段丢失。在星洲银罗非鱼中共发现4种单倍型,单倍型间平均遗传距离为0.008 0,其mtDNA多态度为0.002 4,遗传多样性较丰富。 The authors analyzed restriction-fragment length polymorphism (RFLP) of mtDNA in red tilapia ( Oreochromis niloticus × O. mossambicus ) by 10 restriction endonucleases, EcoR Ⅴ , MboⅡ, Nae Ⅰ, Pvu Ⅱ, Sty Ⅰ, Kpn Ⅰ, Sac Ⅰ, HindⅢ, Dra Ⅰ, Hpa Ⅱ. The result indicated that Sty Ⅰ, Sac Ⅰ and Dra I expressed polymorphism and could be sorted into 4 haplotypes. There were many unolderly cut points by Hpa Ⅱ, which was difficult to analyze; and one cut point by EcoR Ⅴ, Nae Ⅰ, Pvu Ⅱ, Kpn Ⅰ; two cut points by HindⅢ; five points by Mbo Ⅱ but small segments were lost. There were four haplotypes in red tilapia, the average nucleotide diversity of haplotypes is 0.008 0 and the index ofgene diversity among haplotypes is 0.0024, which indicated that there was abundant genetic diversity in ted tilapia
