摘要
目的探讨脑出血模型大鼠脑组织脑红蛋白的表达。方法随机将55只大鼠分为正常对照组(5只)、脑出血组(25只)和假手术组(25只)。采用立体定向自体血注入大鼠尾状核建立脑出血模型(假手术组注入生理盐水),模型建立后2,12,24,48和72h5个时间点分别处死大鼠5只,用RT-PCR方法检测不同时间点脑组织内脑红蛋白的表达。结果RT-PCR和免疫组织化学结果提示脑出血组术后2h脑组织内脑红蛋白相对表达水平开始增高,48h达到高峰,72h开始下降,均明显高于假手术组和正常对照组(P<0.01或P<0.05)。结论脑出血后脑内脑红蛋白表达上调,提示脑红蛋白可能对脑出血继发神经损伤起重要的保护作用。
Objective To investigate dynamic changes of neuroglobin expression in brain tissues of rats with intracerebral hemorrhage. Methods 55 rats were randomly divided into normal control group(n = 5), sham- operated group (n = 25) and intracerebral hemorrhage group (n = 25). The sham- operated group and intracerebral hemorrhage group were divided into 5 subgroups respectively at 2, 12, 24, 48 and 72 hours after the establishment of animal models, every subgroup included 5 rats. Intracerebral hemorrhage was induced by stereotaxic injection of autologous blood into right caudate nucleus of the rats (normal saline was injected in sham- operated group). Both RT - PCR and immunohistochemistry were used to detect the expression of neuroglobin in brain tissues at different times after intracerebral hemorrhage. Results RT- PCR and immunohistochemistry analysis showed that the relative expression level of neuroglobin in brain tissue in the intracerebral hemorrhage group began to increase at 2 hour, reached its peak value at 48 hours, and began to decrease at 72 hours, which were all obviously higher than those in the shamoperated group and that in the normal control group. Conclusion The expression of neuroglobin in brain tissue is upregulated after intracerebral hemorrhage, it is indicated that neuroglobin may play an important protective effect against secondary neuronal injury following intracerebral hemorrhage.
