摘要
①目的探讨卵巢癌流式细胞DNA倍体和S期比例的异质性。②方法应用流式细胞术(FCM)测定了27例卵巢癌的DNA倍体和S期比例(SPF),每例测定2~6块,取自原发癌和腹腔转移癌的不同区域石蜡包埋肿瘤组织。③结果21例DNA指数一致,6例显示DNA指数异质性;DNA异倍体肿瘤中只有9例可进行SPF计算,结果同一病人不同区域SPF变异很大,其中位数为8%(2%~15%);所有DNA二倍体肿瘤的SPF变异中位数为3%(0~7%)。④结论为保证流式细胞DNA倍体及SPF测定能代表卵巢癌的全部细胞干系,应采取多区域取材检测。
Objective To study the flow cytometric DNA ploidy and S phase heterogeneity in ovarian cancer. Methods DNA ploidy and S phase fraction(SPF) from 27 patients with epithelial ovarian carcinoma were analyzed by flow cytometry(FCM).Two to six paraffin embedded tissue blocks from each primary tumor and its metastatic foci were obtained. Results Stable DNA index (DI) was found in 21 patients, heterogeneity was found in 6 patients. SPF measurements were possible only in 9 cases of DNA aneuploid and whose SPF value varied considerably among different samples from the same patient. The median SPF value was 8% (2%~15%). The median SPF value in all DNA diploid cases was 3% (0%~7%) . Conclusion To ensure that all tumor stem lines are represented, multiple biopsy specimens from the primary tumor and its metastatic foci should be analyzed by flow cytometry and SPF measurement.
