乙型肝炎表面抗原破坏试验中对结果影响因素的实验观察 被引量:2

摘要 对二氯异氰尿酸钠和过氧乙酸作用后的纯化HBsAg,以SPRIA法检测结果表明,在其它条件一致时,HBsAg与消毒剂的容量比为1:4或1:9时,检测结果较一致。试剂灵敏度较低,测得消毒剂对HBsAg破坏的作用时间较长。中和剂内小牛血清含量大于5%时,测得消毒合格所需作用时间较不含血清者长。溶液pH值为4.0~9.6对HBsAg抗原性的破坏无影响。 Using purified HBsAg as the marker and sodium dichloroisocyanurate and peracetic acid as the disinfectants, the influences of volume ratio of HBsAg suspension to disinfectant solution, sensitivity of reagent, calf serum content of neutralizer and pH value of the solution on destruction of HBsAg antigenicity were evaluated by SPRIA. The results indicated that when the other conditions are same and the volume ratio of HBsAg suspension to disinfectant solution was 1∶1, 1∶4, 1∶9 and 1∶19, the contact time required by sodium dichloroisocyanurate (containing available chlorine 500 mg/L) or 0.5% peracetic acid for destroying HBsAg antigenicity was 30, 15, 15 and 10 minutes respectively. When the sensitivity of the reagent was low, the contact time required for destroying HBsAg antigenicity was long under the same test conditions. When the calf serum content in the neutralizer was higher than 5%, the contact time required for eligible disinfection was longer than with neutralizer without serum. When the pH value of the solution was 4.0~9.6, there was no influence on destruction of HBsAg antigenicity.
