荧光分光光度法测定血清精氨酸代琥珀酸合成酶

摘要 采用荧光分光光度法测定血清精氨酸代琥珀酸合成酶(ASS)的活力,并对方法的可靠性作了一系列的研究,包括反应产物量与荧光强度,酶量与荧光强度,精密度,敏感度等,结果表明,此法简单、灵敏、可靠。还测定了145例健康成人血清ASS的活力,并对67例肝炎病人血清ASS活力与正常人作了比较,结果显示,肝炎病人组血清ASS活力均显著高于正常成人组(P<0001)。 ? The method of determination of argininosuccinate synthetase(ASS) activity in human serum with fluorescence spectrophotometry was established. The reliability of the method was studied. The relation between the products and fluorescence intensity, between the activity of ASS and fluoresence intensity, the precision, sensitivity,were studied respectively. The results showed that it was a simple, sensitive and reliable analytical method for clinincal diagnosis. The serum ASS activity in 145 heathy adults and 67 hepatitis patients were measured. Normal reference values were established. The results reveal that ASS activity in hepatitis patients was higher than that in normal adults. There was a significant difference(P<0.001) between them.
