摘要
以马鹿为试验对象,采用上浮法诱导马鹿精子体外获能,以孕酮(P4)诱导顶体反应,旨在摸索出P4诱发马鹿精子顶体反应的最适宜体系。研究结果表明:0.5~1000μmol/L的P4可提高获能马鹿精子的顶体反应率;其中,1μmol/L与10μmol/L的P4可显著(P<0.05)促进马鹿精子发生顶体反应,并以10μmol/L的P4处理组的顶体反应率最高(74.44±0.71)%。10μmol/L的P4诱导获能马鹿精子顶体反应在10~20min的处理时间内均获得了较高的顶体反应率(均72.41%以上),并以处理10min的顶体反应率最高(78.40%)。P4能促进获能马鹿精子的顶体反应,但不能显著(P>0.05)促进未获能马鹿精子的顶体反应。
Wapiti (Cervus elaphus) sperm was capacitated in vitro through the method of swimming-up and acrosome reaction (AR) was induced by using progesterone (P4) in order to determine the optimal system using P4 during AR. The results showed that: P4 at 0.5 - 1000 μ mol/L could enhance the ratio of AR on the capacitated sperm. Effects of P4 within the range of 1 μ mol/L and 10 μ mol/L were significant (P 〈0.05) on AR and 10 μ mol/L P4 obtained the highest ratio of AR (74.44±0.71)%. Incubated from 10 to 20min by P4 at 10 μ mol/L, the wapiti sperm abtained higher ratio of AR (over 72.41%) and incubated 10min reached the highest ratio (78.40%). Therefore, AR of capacited wapiti sperm, but not uncapacited sperm (P〉0.05) could be induced by P4.
