摘要 [目的]研究玉米皮中水溶性多糖的制备方法和化学结构。[方法]采用单因素试验研究了多糖的最佳提取工艺,采用高效液相色谱、红外光谱、紫外光谱对多糖结构进行了解析。[结果]玉米皮多糖的最佳提取工艺为:料水比1∶30,在100℃提取2次,每次2h。在该提取条件下,100g玉米皮可制备得到6.94g多糖,多糖纯度为88.18%。玉米皮多糖主要由α-D-吡喃葡萄糖组成,其含量为90.8%。此外,玉米皮多糖还含有少量的阿拉伯糖、木糖、半乳糖等单糖以及微量的蛋白质。[结论]玉米皮中水溶性多糖含量较高,有很好的开发利用价值。 [Objective] The research aimed to study the preparation method and structure of water-soluble polysaccharide in corn skin (PCS). [Method] The optimum extracted condition of polysaccharide was studied by using one-factor experiment and the structure of polysaccharide was analyzed by using HPLC, IR spectrum and UV spectrum. [Result] The optimum extracted conditions were that material:water was 1:30, temperature was 100℃, extracted time was 2 h, extracted times was 2 times. From 100 g corn skin, 6.94 g polysaccharide could be obtained according to the above conditions, and the purity was 88.18%. The main sugar of the polysaccharide was α-D-Glup (90.8%), along with small arabinose(Ara), xylose(Xyl), galaetose(Gal )and protein. [Conclusion] Corn skin contained quite a few water-soluble polysaeeharide, so it had high value of use and development.
