摘要
目的探讨中药姜黄素对膀胱肿瘤细胞(T24细胞株)细胞凋亡发生及组织蛋白酶D表达的影响。方法以体外培养T24细胞为研究对象,姜黄素组加入终浓度为1、10、100μmol/L的姜黄素,对照组不加姜黄素,MTT法、TUNEL染色、逆转录-PCR、免疫印迹及图像分析技术检测12、24、48h后各组细胞活性、凋亡发生、组织蛋白酶D转录水平及蛋白表达变化。结果姜黄素组T24细胞生长抑制率随姜黄素浓度增加显著上升,细胞凋亡发生显著升高,组织蛋白酶D蛋白活性及转录水平显著降低。结论姜黄素能抑制T24细胞组织蛋白酶D的转录和蛋白活性。
Objective To investigate the effect and possible mechanisms of curcumin on the cathepsin D expression and cell ap- optosis of T24 cells. Methods In the control group,there was not any curcumin added. In the curcumin group, the contents of curcu- rain added were 1,10,100μmol/L respectively. T24 cells were observed in 12,24 and 48h after curcumin was added. MTT,TUNEL staining, RT-PCR,immunoblot and image analysis technology were used to detect the growth, apoptosis, cathepsin D protein activity and mRNA level of T24 cell. Results In the curcumin group,inhibitor rate and cell apoptosis increased significantly. The activity and mRNA level of cathepsin D decreased significantly. Conclusion Curcumin can inhibit the expression of cathepsin D of T24 cell in the transcriptional and proteinic level.
