摘要
目的通过大鼠后肢慢性静脉高压模型,研究股静脉中细胞间黏附分子-1(ICAM-1)的动态表达情况。方法72只SD大鼠随机分为实验组和对照组,经腹手术环缩大鼠下腔静脉下端及髂总静脉,实验组和对造组内再分为术后当天(0周)、2、4、8、12、16周6组,取大鼠股静脉,免疫组化染色法观察ICAM-1的动态表达。结果ICAM-1阳性面积百分比,实验组与对照组比较在手术当天差异无显著性(P>0.05),第2、4、8和12周比较差异有显著性(P<0.05),16周比较差异无显著性(P>0.05);对照组内,各时间组比较差异无显著性(P>0.05);实验组内,各时间组间比较差异有显著性(P<0.05)。ICAM-1阳性细胞百分比,实验组与对照组在手术当天比较无显著差别,在第2、4、8周差异均有显著性(P<0.05),12周、16周,差异无显著性(P>0.05);对照组内,各时间组比较差异无显著性(P>0.05);实验组内,各组间比较差异有显著性(P<0.05)。结论慢性静脉高压可大鼠股静脉内皮细胞和平滑肌细胞ICAM-1表达增强,从而介导白细胞对静脉壁重塑的影响。
[ Objective] To investigate the expression of ICAM-1 in the femoral vein of a rat model of chronic venous hypertension. [Methods] Seventy-two SD rats were divided randomly into CVH and sham groups. The inferior vein eava and bilateral common iliae vein of the rats in the CVH group were Constricted after a laparotomy. Both the CVH and sham group were divided randomly into 6 subgroups according to the 0wth, 2wth, 4wth, 8wth, 12wth, 16wth after operation. The samples of the femoral veins were harvested and immunohistochemisty method were used to detect the expression of ICAM-1 in the venous wall. [Result]There were no significant differences among all subgroups of the sham group (P 〉0.05) while there were significant differences among all subgroups of the CVH group (P〈0.05). On 2wth, 4wth, 8wth, The percentage of ICAM-1 positive cells was larger significantly in the CVH group than in the sham group. On 0wth, 2wth, 16wth, there was not different significantly between the CVH group and the sham group (P 〉0.05). There were no significant differences among all subgroups of the sham group (P〉0.05) while there were significant differences among all subgroups of the CVH group (P 〈0.05). [Conclusion] The increased ICAM-1 positive expression of the endothelium cell and SMC suggest that inflammation may be involved in the development of venous remodeling.
