摘要 目的研究过氧化氢诱导的胰岛β细胞株(RINm5F细胞株)凋亡中iNOS的活性变化趋势。方法过氧化氢诱导胰岛β细胞凋亡,采用MTT(细胞活性检测)、SRB(活细胞蛋白染色)鉴定细胞凋亡,对凋亡细胞iNOS的活性测定来反应i-NOS的激活程度。结果过氧化氢在0.1 mmol/L,1 h能明显诱导胰岛β细胞的凋亡,其细胞活性和活细胞染色均较对照组明显下降,过氧化氢诱导后iNOS活性提高约42%。结论iNOS的激活可能是过氧化氢诱导的胰岛β细胞凋亡的机制之一。 OBJECTIVE To investigate into the role of iNOS activation in islet β cell apoptosis induced by hydrogen peroxide. METHOD Hydrogen peroxide was used to induce islet β cell apoptosis, and MTr and SRB were used to identify cell apoptosis and determine the degree of iNOS activation. RESULT Under the condition of 0.1 mol/L and 1 hour, Hydrogen peroxide could markedly induce apoptosis, with decreased cell activity and cell staining. After induction, iNOS activity was increased by 42 %. CONCLUSION The activation of iNOS is probably one of the mechanisms for the induction of islet β cell apoplosis by hydrogen peroxide.
