摘要
目的:评价天然羟基磷灰石/壳聚糖复合材料(nature hydroxyaptite/chitosan composite,NHC)的细胞相容性。方法:人骨髓间充质干细胞诱导法培养成骨细胞,并鉴定。将材料与细胞体外进行复合培养,应用细胞增殖度,碱性磷酸酶和扫描电镜等对其细胞相容性进行评定。结果:骨髓间充质干细胞诱导的成骨细胞在复合材料表面粘附,生长良好,细胞增殖度无显著性差异(P>0.05),材料组与对照组之间碱性磷酸酶无显著性差异(P>0.105),扫描电镜观察发现24h后细胞生长良好,细胞连接紧密,有细胞外基质生成。说明本复合材料具有良好的细胞相容性。结论:NHC具有较良好的体外细胞相容性。
Objective To investigate the cellular biocompatibility of nature Hydroxyaptite/ Chitosan composite (NHC). Methods The composite was cocultured with osteoblasts derived from human bone marrow stem cells. The cellular biocompatibility was studied by means of the evaluation of proliferation capacity, alkaline phosphatase staining and scanning electron microscope observation. Results Excellent adherence and growth of osteoblasts were observed on the composite surface. There were no significant differences in both cell proliferation capacity and alkaline phosphatase staining between the experiment and control groups, SEM observation showed normal growth of osteoblasts and extracellular matrix generation in 24 hours. Conclusion NHC had good biocompatibility with the osteoblasts in vitro.
