摘要
目的检测血管活性肠肽(vasoactive intestinal peptide,VIP)在人胃溃疡组织中的表达和分布,探讨VIP在胃溃疡发病机制中的作用,为内分泌治疗提供理论依据。方法收集潍坊医学院附属医院2000-2005年的胃溃疡标本进行免疫组织化学染色,观察VIP在胃溃疡组织中的表达,收集新鲜的胃大部切除手术后胃溃疡标本,采用RT-PCR(re-verse transcription polymerse chain reaction)检测VIP mRNA的表达。结果胃溃疡边缘VIP免疫阳性物质在黏膜层和黏膜下层较正常胃组织显著减少(P<0.01)。胃溃疡VIP免疫阳性物质在肌间神经丛、肌层分布丰富,较正常胃组织明显增多(P<0.05)。RT-PCR结果表明在正常胃组织和胃溃疡组织中VIP均有表达,光密度分析显示在胃溃疡VIP光密度比值大于正常胃组织VIP光密度比值(P<0.05)。结论免疫组织化学及RT-PCR结果在分子和基因水平显示胃溃疡中VIP表达的变化,表明VIP在胃溃疡发病中起着重要的作用。
Objective To investigate the expression and distribution of vasoaetive intestinal peptide(VIP) in human gastric ulcer and its effects in pathogenesis of the gastric ulcer, and provide the theory data for endocrinal therapy. Methods The tissues of the gastric ulcer were collected in the affiliated hospital of Weifang Medical College from 2000 to 2005 and were dyed by immunohistochemistry. The fresh tissues of the gastric ulcer were collected and the expression of VIP mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR). Results The VIP positive materials decreased in the mueosa and the submucosa in the margin of the gastric ulcer compared with the normal tissue ( P〈0.01 ), but increased the muscular intramuscular plexus layer tha, the normal one( P〈0.05 ). The results by RT-PCR indicated the expression of VIP in the normal and the ulcer. The OD ratio was higher in the ulcer tissue than the normal one by the photedensitometry (P〈0.05). Conclusions The results of the immunohistoehemistry and RT-PCR showed the diversity of VIP in the gastric ulcer in the level of molecule and gene, VIP plays an important role in the pathogenesis of the gastric ulcer.
