摘要
以康宁木霉3.4261为出发菌株,分别经紫外线诱变、紫外线与硫酸二乙酯复合诱变,采用刚果红透明圈法初筛和摇瓶复筛,获得1株高产纤维素酶的突变株E20-2,初步确定了固态发酵最佳产酶条件:麦麸︰秸秆=1:4,固:液=1:3~1:4,0.1%Tween-80,温度28℃、pH6.0、时间5d,纤维素酶(CMCase)、滤纸酶(FPA)酶活分别达到654U/g和69U/g,较出发菌株分别提高了230%和72%。
Trichoderma koningii 3.4261 was used as the original strain and treated with UV and EMS for mutation. One high cellulase-pmducing mutant strain named E20-2 was obtained by screening with Congo red transparent zone method and shake flask fermentation. Preliminary analysis showed the optimal fermentation conditions were: wheat bran: wheat straw of 1:4, solid: liquid of 1:3-1:4, Tween-80 concentrations 0.1%, temperature 28℃, pH 6.0, cultured for 5 days. The CMCase and FPAase of the mutant strain reached 654 U/g and 69 U/g, which were 230% and 72% higher than that of the parent strain, respectively. And E20-2 had a relatively higher cellulase producing ability than ever reported.
