精神分裂症患者脑脊液对食蟹猴行为的影响

摘要 目的研究精神分裂症患者脑脊液对食蟹猴行为的影响。方法根据大脑CT检查结果,将入组精神分裂症患者分为脑结构异常组和脑结构正常组,留取脑脊液样本3ml;将食蟹猴随机排序后经腰椎穿刺在其脑脊髓腔内注入两组精神分裂症患者、健康志愿者脑脊液标本或生理盐水,形成实验组I(脑结构异常组脑脊液处理组)、实验组Ⅱ(脑结构正常组脑脊液处理组)、对照组(健康志愿者脑脊液处理组)和生理盐水组,其后行为观察13周,利用自编食蟹猴行为评估量表进行评定,取基线,处理后第2,4,6,8,10,12周末评估数据进行分析。结果(1)处理后实验组I[(31.00±3.83)分]、实验组Ⅱ[(28.20±4.92)分]的行为量表总分较处理前[(21.67±1.94)分,(22.33±0.47)分]增高,对照组处理后[(17.00±4.24)分]较处理前[(22.17±0.24)分]明显下降,生理盐水组在处理前后无明显变化;实验组I、Ⅱ在处理后6个观测点的行为量表总分均高于对照组和生理盐水组;处理前后量表总分的方差分析表明,处理和时间的交互效应具有统计学意义(F=21.67,P〈0.01);(2)处理后各观测点8个行为指标的秩和检验表明,实验组I、Ⅱ的行为改变以躁动(P=0.026)、攻击性(P=0.046)、一般活动(P=0.039)三个因子分增高为主,并随时间而明显。结论将精神分裂症患者脑脊液注射入食蟹猴脑脊髓腔可使食蟹猴出现活动增加、攻击性增强、躁动等行为异常。 Objective To investigate whether there were behavioral changes of macaque after being injected cerebrospinal fluid (CSF) of schizophrenics in their cerebrospinal cavities. Methods By random selection strategy, according to the cerebral Computer Tomography (CT) presentations, CSF of schizophrenics with abnormal brain structures, schizophrenics with normal brain structures, healthy volunteers, and normal saline were separately injected into macaque's cerebrospinal cavities via lumbar puncture. According to the inoculated matter, macaque were divided in sequence into case group A (CGA) ,case group B (CGB) ,healthy control group(HCG) and normal saline group(NSG). Behavioral scale made by investigators themselves was used for assessing macaque' s behaviors for 3 weeks as the basis data before inoculation and for 12 weeks after that. Data of basis,the 2nd,4th, 6th ,8th, 10th, 12th weekend after treatments were used in statistic analysis. Results After treatments the total behavior scale scores of CGA (31.00 ± 3.83 ) and CGB (28.20 ± 4.92 ) were higher than that before treatments (21.67 ± 1.94,22.33 ± 0.47 respectively). HCG got lower total behavior scale score after treatment (17.00 ± 4.24) than before (22.17 ±0.24). The total behavior scale score of NSG after treatment was not different from it before treatment, Two case groups have higher total scores of behavior scale than HCG and NSG at 6 time point after treat. General linear model analysis show treatment * time intercept effect ( F= 21.67, P〈0.01 ) were statistically significant. Scores of 8 behavioral factors at every time points after treatments were analyzed by Wilcoxon test. The results show all two case groups had higher scores of aggressiveness (P=0.046) ,general activities ( P = 0. 039) , feistiness ( P = 0. 026) than HCG and NSG, and these differences got more significant with time. Conclusion Injecting schizophrenic CSF into macaque' s cerebrospinal cavities may result in significant behavioral changes of macaque, such as more aggressive, activating and feisty.
