肺移植中应用雷公藤内酯醇保护供肺的实验研究 被引量:1

摘要 目的:探讨雷公藤内酯醇(TRI)在兔肺移植中对供肺缺血再灌注肺的保护作用及可能的作用机制。方法:将16对健康家兔随机分为两组,对照组肺以改良低钾右旋糖酐(LPD)肺保护液灌注及保存,实验组则将雷公藤内酯醇(0.5 mg/L)加入改良LPD液灌注及保存,在4℃保存4 h后再灌注1 h,测定肺组织中核转录因子-κB(NF-κB)的表达以及细胞间黏附分子-1(ICAM-1)的表达;并观察肺组织病理结构的变化。结果:再灌注后,实验组的肺组织中NF-κB和I-CAM-1的表达明显低于对照组(P<0.01),肺组织病理变化较对照组轻。结论:雷公藤内酯醇能减轻肺的缺血再灌注损伤,对供肺具有明显的保护作用。 Objective:To investigate the protective effects of triptolide(TRI)on ischemia-reperfusion during rabbit lung-transplant, and explore its possible mechanism. Meth'ods:Sixteen pairs of rabbit were randomly divided into two groups : the control group and the trail group. The harvested lung blocks were flushed with and stored in the modified low-postasssium-dextron (LPD) solution in control group ,but in the trail group LPD containing triptolide (0. 5 mg/L) was used. The lungs were stored at 4 C for 4 h in both groups. The rabbit lungltransplant reperfusion models were es- tablished and the lungs were perfused for 1 h. After reperfusion,the lung tissue protein expression of nuclear factor-kappa B (NF-κB) and intercellular adhesion molecule-1 (ICAM-1)were also detected at the end of reperfusion. Light microscopic changes were also examined. Results:After reperfusion the protein expression of NF-κB and ICAM-1 were decreased evidently in trail group (P〈0. 01). The morphologic and ultrastructual damages in trail group were milder than in control group. Conclusion:Triptolide can alleviate the ischemia-reperfusion injury of lung and improve the protective effect of lung preservation.
