摘要
目的研究小菜蛾羧酸酯酶的性质.方法CarE活性测定是以α-NA或β-NA为底物(含毒扁豆碱10-5mol/L),在30℃下水浴反应15分钟,然后加入1 ml显色液终止反应,分别在555 nm和600 nm处测定光密度值.结果小菜蛾羧酸酯酶(CarE)性质研究表明:抗性种群和敏感品系成虫CarE活性均以2日龄为最高,4日龄时活性最低;武汉和张家口种群幼虫CarE对α-NA和β-NA的水解活性明显高于敏感品系,但是成虫CarE活性在三个种群间没有明显差异;张家口种群对α-NA和对β-NA的亲和力都明显低于其它田间抗性种群,武汉、宣化、北京三个种群对α-NA或对β-NA的亲和力近似.结论本研究为小菜蛾的抗药性治理奠定了生物化学基础.
Objective To study characters of Care of Plutella xylostella. Methods Substrate of men- surating activity of Care was with α-NA or β-NA, responsed in water for 15 minutes at 30℃, then joined 1ml Obvious liquid, mensurated optic density at 550. nm and 600nm. Results The characters of Care of Plutella xylostella were studied; the results showed that activities of Care different population were the highest in the secondary day and were the lowest in the fourth day. The hydrolyzing characters of Care toward ATCH of larvae of Wuhan population and Zhangjiakou population were highter than sensitivity population, but characters of adults there were not obviously different with other populations. The affinity of Care toward ATCH of Zhangjiakou population was obviously lower than others. Three chemical resistance populations of Wuhan, Xuanhua and Beijing, were near to the level. Conclusion' This study has laid the biochemical foundation for treatment chemical resistance of Plutella xylostella .
