目的通过检测Survivin基因在喉癌及其相关组织的表达,了解其在喉癌发生发展中的作用。方法应用反转录-聚合酶链反应(RT-PCR)检测喉鳞癌组织、癌旁组织、非典型增生组织和正常对照组织中Survivin mRNA的表达水平。结果Survivin mRNA在喉癌的表达与其他非癌组织中的表达差异有统计学意义(P<0.01),其在喉癌的表达与喉癌组织分化程度、TNM分期及淋巴结转移均无明显相关关系(P>0.05)。结论Survivin在喉癌中高表达可能分别是凋亡调控的关键事件,与喉癌的发生发展有关。
Objective To find whether there is difference in survivin level between laryngeal neoplastic tissues and laryngeal non- neoplastic tissues, and to explore the expression of survivin in normal mueosa, dysplasia, squamous cell carcinomas and adjacent non-neoplastic tissues of the larynx to search for clinical usefulness. Methods The mRNA expression of the survivin gene was evaluated by reverse transcription-polymerase chain reaction (RT-PCR) in 40 laryngeal carcinomas and in 52 non-neoplastic laryngeal tissues. Results The mRNA expression levels of Survivin was increased in the transition from normal mucosa, adjacent non-neoplastic tissues to laryngeal neoplastic tissues (P〈 0.01). Conehtsion Compared with non-neoplastic laryngeal tissues, survivin was overexpressed in laryngeal squamous cell carcinomas, suggesting that regulation of apoptosis might be involved in laryngeal neoplasm. The patterns of expression observed for survivin in non-neoplastic laryngeal tissues suggest that the control of apoptosis might also be involved in normal larynx mucosa development.
