眼镜蛇毒因子在大鼠脑缺血-再灌注损伤中的作用研究

摘要 目的探讨眼镜蛇毒因子(CVF)抑制补体在大鼠脑缺血-再灌注损伤中的作用。方法采用线栓法制备大鼠大脑中动脉闭塞(MCAO)模型,缺血2h后再灌注。治疗组用CVF抑制补体腹腔内注射,对照组注射等量生理盐水,检测缺血-再灌注后不同时点各组脑组织补体C1q和C3d蛋白表达情况及神经功能缺损评分、脑组织含水量的变化。结果治疗组补体C1q和C3d表达、神经功能评分及脑组织含水量均明显低于对照组(均P<0.05)。结论CVF具有脑保护作用。 Objective To investigate the inhibited complement effect of cobra venom factor(CVF) on the cerebral ischemic-reperfusion injury of rats. Methods The middle cerebral artery occlusion(MACO) model was induced through filament method. Rats were subjected of ischemia for 2 h by MCAO and then reperfused. After ischemia the line was extracted. Treatment animals were injected with CVF intraperitoneally. Control rats received an equal volume of saline injected intraperitoneally. The dynamic changes of the markers were estimated such as the expression intensity of complement C1q and C3d protein, brain water content and the change of the neurologic impairment score. Results CVF could significantly inhibit the expression of complement C1q and C3d, alleviate brain water content(P〈0.05)and improve the neurologic impairment.Conclusions CVF had neuroprotective strategy.
