艾灸治疗大鼠糖尿病周围神经病变的周围神经保护机制 被引量:31

Mechanism of Peripheral Neuroprotection Induced by Moxibustion Treatment of Rat Diabetic Peripheral Neuropathy

摘要 目的探讨艾灸治疗糖尿病周围神经病变(diabetic peripheral neuropathy,DPN)的周围神经保护机制。方法以链脲佐菌素(Streptozotocin,STZ)腹腔注射诱导形成DPN模型,艾灸大鼠"胰俞"、"足三里"穴,每日1次,每穴15min,连续治疗56天。通过血糖、尿糖、体重、饮食量等的监测,结合神经电生理方法检测神经传导速度,评价艾灸对DPN的治疗效应;HE染色、光镜观察坐骨神经组织结构变化;酶联免疫吸附法检测坐骨神经NGF含量。结果治疗后,艾灸组大鼠血糖水平明显低于模型组(P<0.01);艾灸组感觉神经传导速度明显快于模型组(P<0.01);艾灸组病理形态较模型组有改善;艾灸组NGF含量也明显高于模型组(P<0.01)。结论1.艾灸能有效改善DPN模型大鼠周围神经病变症状、体征。2.艾灸改善DPN模型大鼠周围神经症状的效应可能与提高NGF含量,促进周围神经保护有关。 Objective To investigate the mechanism of peripheral neuroprotection induced by moxibustion treatment of diabetic peripheral neuropathy(DPN).Methods A DPN model was made by intraperitoneal injection of Streptozotocin(STZ).Rat points Yishu(Ex-B_3)and Zusanli(ST 36)were moxaed 15 min each,once daily for 56 consecutive days.The therapeutic effect of moxibustion on DPN was evaluated by measuring blood sugar,urine sugar,body weight and food intake and determining nerve conduction velocity through a neuroelectrophysiological method.Changes in the histological structure of the sciatic nerve were observed optical-microscopically by HE staining.The NGF content of the sciatic nerve was measure by enzyme linked immunosorbent assay.Results The level of blood sugar significantly decreased(P〈0.01),nerve conduction velocity significantly increased(P〈0.01),pathomorphology significantly changed(P〈0.01)and the NGF content significantly increased(P〈0.01)in the moxibustion group after treatment compared with the model group.Conclusion 1.Moxibustion can effectively improve the symptoms and signs of diabetic peripheral neuropathy in a rat model of DPN.2.The improving effect of moxibustion on diabetic peripheral neurological symptoms in a rat model of DPN may be related to an increase in the NGF content and promotion of peripheral neuroprotection.
