摘要
通过自花和异花授粉、电镜扫描、花粉活力测定等方法,对转pCAMBIA1301基因蓝猪耳的9个转基因株系的F1代植株的育性进行了研究.结果表明:蓝猪耳不存在自交不亲和现象但其转基因植株出现花粉败育;对9个转基因株系进行测定,发现其花粉活力与对照(77.3%)差异显著,仅为9.8%- 30.6%,表明转基因蓝猪耳花粉败育不是由于基因的插入引起的,普通培养基组培瓶蓝猪耳花药活力与在土壤中萌发的蓝猪耳花粉活力均为75%左右,表明组培环境也不是引起花粉败育的原因,因此继代培养基是引起花粉不育的唯一原因.
The fertility of F1 plants from 9 transgenic plant lines of Torenia fournieri with transgenic vector pCAMBIA1301 were studied by artificial pollination experiments, SEM observation and pollen rigour test. The results showed that no self-incompatibility appeared in artificial self pollination of Torenia fournieri plants, but pollen abortion had occurred in its transgenic plants. According to pollen vigour test, pollen vigor of 9 transgenic plant lines ranged between 9.8 0% and 30.6 0%, significantly lower than that (reached 77.3 %)of the control,which indicated pollen sterility was not the result of inserted genes. In addition,pollen vigor of plants both cultured in general media of the pots and originated from seed germination in the soil of greenhouse reached about 75 % ,and it was proved that the reason of pollen abortion of transgenic plants of Torenia fournieri was actually not the tissue culture media, but was the subculture media.
