含何首乌血清对人胚肺二倍体成纤维细胞端粒酶活性的影响 被引量:3

摘要 目的观察含何首乌血清对人胚肺二倍体成纤维细胞(2BS)超氧化物歧化酶(SOD)活性、c-myc mRNA表达及端粒活性的影响。方法采用含何首乌血清对2BS细胞株进行处理,用黄嘌呤氧化酶法检测SOD活性,RT-PCR检测c-myc mRNA表达,TRAP-ELISA检测端粒酶活性。结果与正常对照组相比,用药组2BS细胞SOD活性明显增加(P<0.05),c-myc mRNA表达无变化,端粒酶活性显著增加(P均<0.05)。结论含何首乌血清对2BS细胞具有抗衰老作用,其机理可能与促进自由基的清除和激活端粒酶活性有关。 Objective To observe effects of Tuber Fleeceflower Root serum on activity of SOD, the expression of c- myc mRNA and the activity of telomerase of human fetal lung diploid fibroblasts (2BS). Methods 2BS cell lines were treated with Tuber Fleeceflower Root serum. The activity of c- myc rnRNA was detected by semi - quantitive RT- PCR. A TRAP- ELISA method was used to measure the telomerase activity. Results Compared with those in the control group, the activity of SOD increased obviously ( P 〈 0.05), the expression of c - myc mRNA remained on the same level, and the telomerae activity were enhanced significantly( P 〈 0.05) in the drug group. Conclusion Tuber Fleeceflower Root serum can postpone senescence of 2BS, its mechanism may be associated with enhancement elimination of free radicals and activation of telomerase.
