rhEGF对人皮肤成纤维细胞的促增殖作用 被引量:5

摘要 目的研究重组人表皮生长因子(rhEGF)在不同浓度下对人皮肤成纤维细胞增殖的影响,为临床准确定量应用这两种因子修复创面、加速创面愈合提供理论依据。方法体外培养人皮肤成纤维细胞,分别用浓度为0,10,30,50,80,100,150μg/L的rhEGF干预细胞,48h后四甲基偶氮唑盐比色法(MTT)检测细胞的活性。选择最适浓度生长因子干预的细胞,用流式细胞仪检测细胞的DNA倍体情况。结果不同浓度的rhEGF分别对成纤维细胞干预48h后,显示rhEGF在80μg/L浓度时MTT值最大;并检测出在最适浓度生长因子干预24h后细胞大多处于DNA合成前期和合成期。结论rhEGF浓度在30-100μg/L之间对人皮肤成纤维细胞有明显的促增殖作用,其中以80μg/L较显著。 Objective To investigate the effect of different concentrations of rhEGF on human skin fibroblasts. Methods The fibroblasts were cultured in vitro and interfered with different concentrations of rhEGF. After 48 h, cell activity was detected with MTT. The DNA ploidy of ceils interfered with the best concentration were detected with FCM. Results MTT value was highest after interfered with 80μg/L rhEGF. The ceils interfered with optimum concentration for 24 h were in pre- and synthetic DNA synthesis period. Conclusion RhEGF at the concentration of 80μg/L is more effective to induce the proliferation of fibroblasts.
