摘要
目的了解焦炉工人外周血淋巴细胞抑癌基因p53突变情况,探讨外周血淋巴细胞p53基因外显子5、6、7突变规律与焦炉暴露的关系。方法选取焦化厂焦炉作业工人93名,按不同工种分为炉底组(n=33)、炉侧组(n=30)、炉顶组(n=30),无任何PAHs接触史的库工84名作为对照组。对作业环境进行监测;取受试者的班末尿和肘静脉血,用高效液相色谱法检测尿中1-羟基芘的水平,用聚合酶链式反应-单链构象多态性方法(PCR-SSCP)检测外周血淋巴细胞p53基因突变的情况。结果焦炉工人暴露组血淋巴细胞p53基因突变率(32.2%)显著高于对照组(15.5%,P<0.05),且血淋巴细胞p53基因突变率随外暴露水平升高(对照组<炉底组<炉侧组<炉顶组)而升高(χ2=8.831,P<0.01)。p53基因的第6、7外显子突变率呈现随外暴露水平升高(对照组<炉底组<炉侧组<炉顶组)而升高的趋势(χ2=8.556,9.368,P<0.01)。结论焦炉逸散物可诱发焦炉工人的血淋巴细胞p53基因突变率升高,p53基因的第6、7外显子对焦炉逸散物暴露更为敏感。
Objective To investigate the mutations of p53 gene in oven workers' peripheral blood lymphocyte, and to explore the relationship between the mutation at exon 5,6,7 of p53 gene and exposure to polycyclic aromatic hydrocarbons(PAHs). Methods Ninety-three coke oven workers in steel factory were divided into three groups:oven top group ( n = 33 ), oven side group ( n = 30), oven bottom group( n = 30). Eighty-four workers unexposed to occupational carcinogen were selected at the warehouse of steel factory as control group. The operating environment was monitored. Blood and urine samples were collected from 93 coke oven workers and 84 control workers after the night shift. The concentration of urinary 1-hydroxypyrene(HpU)was measured using high performance liquid chromatography(HPLC), and the p53 gene mutation in peripheral blood lymphocyte was detected by polymerase chain.reaction single strand conformation polymorphism analysis(PCR-S&P). Results The p53 gene mutation rate in exposure groups(32.2% ) was significantly higher than in control group(15.5%, P〈0.05). The p53 gene mutation rates in peripheral blood lymphocyte showed a tendency of top-oven workers〉side-oven workers〉 bottom-oven workers〉 controls(x^2 = 8. 831, P 〈 0.01 ). And the p53 mutation rates of the exon 6 and 7 had the same trend(x^2 = 8. 556 and 9. 368, P 〈 0.01, respectively). Conclusion The p53 gene mutation in peripheral blood lymphocyte could be induced by exposure to coke oven emissions(COE), and exons 6,7 are more sensitive to COE.
