摘要
目的探讨转移相关蛋白(metastasis associated protein-1,Mta-1)和血管内皮生长因子(vascular endothelial growth fac-tor,VEGF)在膀胱移行细胞癌(bladder transitional cell carcinoma,BTCC)发生发展过程中的作用及其临床价值。方法免疫组化方法检测40例BTCC组织和10例正常膀胱组织中转移相关基因(Mta-1)及血管内皮生长因子(VEGF)的表达;分析Mta-1与BTCC的分级、分期、侵袭转移及复发间的关系及Mta-1表达与VEGF表达的相关性。结果BTCC中Mta-1蛋白表达阳性率为70.0%(28/40),明显高于正常膀胱组织(0,P<0.05);Mta-1蛋白表达阳性率随BTCC临床分期及病理分级的升高而增加,肿瘤复发组高于初发组,肿瘤转移组高于无转移组(P<0.05)。VEGF的表达在BTCC组明显高于正常对照组(P<0.05),BTCC组织中VEGF表达阳性率随肿瘤临床分期的升高及复发、转移的发生而升高。BTCC中VEGF蛋白表达与Mta-1蛋白表达呈正相关(rs=0.762,P<0.05)。结论结果提示Mta-1和VEGF表达与肿瘤的转移及复发密切相关,可以为膀胱癌基因治疗和抗肿瘤血管形成治疗提供新靶点。
Objective To explore the role of metastasis associated protein-1 (Mta-1 )and vascular endothelial growth factor(VEGF) expression in the occurrence and development of BTCC and their clinical value. Methods In 40 cases of BTCC and 10 cases of normal bladder tissues, the expression of Mta-1 and VEGF was examined with immunohistochemical method, and their correlation was analyzed. Their correlation with the invasion, metastasis, angiogenesis and recurrence of BTCCs was analyzed. Results The positive rates of Mta-1 expression were 70.0 % (28/40) in BTCCs and 0 in normal bladder mucosa tissues( P 〈 0.05 ). The expression level of Mta-1 was higher in the higher clinical stages and pathological grades of BTCCs, and it was higher in recurrence group than in non-recurrence group, in metastasis group than in non-metastasis group(P〈 0.05). The expression level of VEGF was higher in the higher clinical stages of BTCCs. The positive rate of VEGF expression was significantly higher in BTCCs than that in normal bladder mucosa tissues(P〈 0.05). Mta-1 expression was positively correlated with VEGF expression in BTCCs( rs = 0. 762, P 〈 0.05 ). Conclusion The results indicate that Mta-1 and VEGF could be new targets for gene therapy of bladder neoplasms and anti-neovascularization.
