复发性涎腺腺样囊性癌裸鼠移植瘤模型的建立及其生物学特性 被引量:2

摘要 目的建立人复发性涎腺腺样囊性癌裸鼠移植瘤动物模型。方法将人复发性涎腺腺样囊性癌组织块,进行裸鼠皮下接种、传代,建立裸鼠移植瘤模型,并对其进行光镜、电镜、免疫组织化学及染色体分析。结果人复发性腺样囊性癌组织块移植于裸鼠后,获原代移植成功。2年期间移植瘤在裸鼠之间共连续传代4代,移植BALB/C裸鼠5只,平均潜伏期44.33天。经光镜、电镜、免疫组织化学观察发现移植瘤与其原发肿瘤结构特征一致。染色体检查为亚中着丝点染色体,符合人类体细胞核型。结论成功建立人复发性涎腺腺样囊性癌裸鼠移植瘤模型并传代,移植瘤保留了原发肿瘤的组织学特征和染色体核型。 Objective To establish a model of recurrent adenoid cystic carcinoma (RACC) in nude mice, and to identify the characterization of the transplanted tumor. Methods Tumor tissue of RACC was implanted a nude mice subcutaneously. A series of studies had been performed to observe the morphologic changes by hght microscope ( LM ), transmission electron microscope (TEM), and immunohistochemical stains against CK8 and smooth muscle actin. Chromosome analysis was also done to identify the characterization of the transplanted tumor. Results A model of human RACC transplanted to nude mouse was successfully established and had been successfully passgaed 4 generations in 5 nude mice during two years. The average latent period of the transplanted tumor was 44.33 days. The original morphological and biological characteristics of the tumor were still preserved in the transplanted tumor. The chromosome of the transplanted tumor was subtelocentric which exhibited the features of human cancer. Conclusion The model of RACC in nude mice had been established and the transplanted tumor maintained the properties of human ACC.
