氯沙坦对大鼠肾脏缺血再灌注足细胞损伤保护作用的实验研究 被引量:2

摘要 目的探讨氯沙坦对大鼠肾脏缺血再灌注足细胞损伤的保护作用。方法将健康雄性SD大鼠随机分为4组:假手术组、缺血组、缺血再灌注组、氯沙坦治疗组。处死动物,留取肾组织行电镜观察足细胞损伤,免疫荧光及Western blot检测Nephrin表达变化。结果缺血再灌注组出现足细胞足突融合,部分脱落,Nephrin表达明显减少,氯沙坦治疗组足细胞损伤减轻,Nephrin表达有所恢复。结论血管紧张素Ⅱ部分介导了肾脏缺血再灌注中足细胞的损伤,给予其受体阻断剂可起到一定的足细胞保护作用。 Objective To investigate the protective effects of losartan against podoeyte impairment after I/R. Methods Twenty-four SD rats were randomized into 4 groups: sham operation group, ischemic group, ischemia/reperfusion (I/R) group and losartan-treated group. Podocyte injury was studied with electron microscope, and the expression of nephrin in kidney was examined using immunofluoreseent assay and Western-blotting. Results The,rats in I/R group presented with reduced nephrin expression, foot process effacement and loss following ischemic injury. These effects were considerably ameliorated with Losartan treatment. Conclusion Angiotensin receptor blocker (ARB), such as losartan, may be protective against podocyte impairment following ischemia/reperfusion injury, which is partly mediated by angiotensin Ⅱ.
