摘要 目的:探讨Hepsin蛋白对肝癌细胞生长和细胞凋亡的影响。方法:运用RT-PCR技术检测肝癌组织样品肝癌细胞系中Hepsin基因的表达水平,并从原发性肝癌患者的癌组织中扩增Hepsin基因编码区序列,该序列被克隆并构建pCMV-tag-HS表达质粒。将pCMV-tag-HS表达质粒转染人肝癌细胞系HepG2细胞、Bel-7402细胞,观察Hepsin蛋白对肝癌细胞的增殖能力和细胞凋亡效应的影响。结果:Hepsin基因在临床肝癌组织和人正常肝细胞L-02细胞中高表达,在人肝癌细胞系Bel-7402细胞、HepG2细胞和临床肝癌癌旁组织不表达或低表达,Hepsin蛋白可以增强肝癌细胞系的增殖能力,同时抑制肝癌细胞凋亡。结论:Hepsin蛋白促进肝癌细胞生长并抑制肝癌细胞凋亡。 Objective: To investigate the effect of Hepsin protein on the proliferation and apoptosis of hepatic cancer cells (HCCs). Methods: The expression of Hepsin gene in HCCs and hepatoma cells were determined by RT-POR, and entire ORF of Hepsin was isolated from the products of RT-PCR. Hepsin gene was inserted into expressing vector pCMV-tag-HS. The recombinant vector pCMV-tag-HS was transfected into liver tumor cell line Hep G2 and Bel-7402. Effects of expression of Hepsin on cell growth and apoptosis of hepatoma cell line Bel-7402 and HepG2 were evaluated. Results: Hepsin gene was highly expressed in liver cancer and normal liver cells, and was not expressed in hepatoma cells. The expression of Hepsin facilitated liver cancer growth and inhibited the apoptosis of cell line Hep G2 and Bel-7402. Conclusion: The expression of Hepsin could enhance the proliferation and inhibit the apoptosis of human hepatoma cells.
