摘要
采用细胞因子活性检测,Northernblot及RT-PCR等方法,在体外研究了抗细菌核心糖脂域McAb(EL1、EL3和3H4)对LPS诱导人外周血单核细胞(hPBMC)释放IL-1及其mR-NA表达水平的影响。结果表明,EL1、EL3和3H4在体外有抑制LPS诱导细胞释放IL-1的作用;Northernblot及RT-PCR结果证实,这3株McAb能降低IL-1mRNA表达的水平;提示抗细菌核心糖脂域McAb可通过与LPS的结合而中和LPS作用于效应细胞,从而降低或阻碍了效应细胞中IL-1mRNA的表达,达到抑制细胞释放IL-1的作用。
The effects of monoclonal antibodies(McAb) to core glycolipid(CGL)region of lipopolysaccharides(LPS) on the dynamic change and mRNA expression of Interleukin 1(IL 1)in supernatants of cultured human peripheral blood monocyte(hPBMC) induced by LPS in vitro were studied by cytokine bioassay,Northern blot and RT PCR analyses.The results showed that the IL 1 levels in supernatants of hPBMC were reduced in groups treated with anti CGL McAbs (EL1,EL3 and 3H4).Northern blot and RT PCR analyses showed that mRNA expression of IL 1 in hPBMC after LPS induced were also inhibited by these three anti CGL McAbs.These evidences suggest that anti CGL McAbs are able to influence mRNA expressions of LPS induced cytokines by blockade of the combination of LPS with effector cell,and consequently reduce its production.
