Cellular Immune Responses to Humans by Inoculation of Freeze-dried Live Attenuated Hepatitis A Vaccine(H_2 Strain)

摘要 目的了解接种冻干甲型肝炎(甲肝)减毒活疫苗[Hepatitis A(Live)Vaccine,Freezedried;Hep A]后,机体能否产生特异性细胞免疫应答。方法研究对象经HepA(H2株)1剂次免疫,流式法检测淋巴细胞亚群CD2^+、CD4^+、CD8^+的百分率及表达干扰素(IFN)-γ及白细胞介素(IL)-4的阳性细胞百分率;T淋巴细胞的特异性增殖试验(MTT法)检测T淋巴细胞增殖反应;酶联免疫斑点法(ELISPOT)检测淋巴细胞的IFNγ分泌能力。结果接种后2周,CD^+上升,以4周上升为显著,与接种前相比,差异有显著的统计学意义(t=2.571,P〈0.05);接种后4周~3个月,淋巴细胞对特异性刺激物甲肝病毒的反应都有明显增强,与接种前相比,差异均有非常显著的统计学意义(t=3.215~3.959,P均〈0.01);接种后2周,表达IL-4和IFN-γ的阳性细胞百分率上升,以IL-4为明显,与接种前相比,差异有显著的统计学意义(t=2.197,P〈0.05);接种后3年,用ELISPOT法检测IFN-γ,阳性率为57.1%(8/14)。结论接种HepA(H2株)后,机体能有效地产生特异性细朐免疫席答。 Objective To study the specific cellular immune responses of humans induced by Freeze-dried Live Attenuated Hepatitis A Vaccine(H2 Strain). Methods Sixteen subjects,who had undetected anti-HAV antibody, were inoculated with one dose Freeze-dried Live Attenuated Hepatitis A Vaccine(H2 Strain). T lymphocyte sub-cluster CD3^+ , CDf , CD8^+ and the expressing cells of IFN-γ,IL-4 were tested by FCM. Lymphoproliferative responses were measured by MTT assay. IFN-γ-producing cells were detected by ELISPOT assay. Results Compared with prevaccination,the number of CD4^+ increased at two weeks after vaccination,then reached peak level at four weeks (t=2. 571 ,P 〈 0. 05); Lymphoproliferative responses to HAV was obviously increased at 4 weeks to 3 months (t =3. 215 ~ 3. 959,P 〈 0.01) ; The levels of ceils of both IFN- γ, and IL-4 increased at two weeks after vaccination,which the increasing of IL-4-expressing cells was higher than that of IFN-γ cells. The difference has obvious statistic signification (t=2. 197, P〈0.05). At 3 years after vaccination, posttive of IFN-γ-producing cells were 57.1% by ELISPOT assay. Conclusion The can elicit Specific cellular immune responses can be produced after administrating Freeze -dried Live Attenuated Hepatitis A Vaccine (H2 Strain).
