摘要 目的:观察利用高效液相色谱-紫外法检测尿液8-羟基脱氧鸟苷(8-OHdG)的效果。方法:尿液经前处理后,利用美国Agilent高效液相色谱仪检测8-OH-dG水平。结果:8-OHdG峰与杂质峰分离良好,8-OHdG浓度在0.5~20μg/ml之间呈良好线性,流动相最佳的pH为3.5,回收率在98.6%~100.1%之间,批内差异(CV)均低于8%。结论:高效液相色谱-紫外法是一种灵敏度较高、重复性好、易推广的尿液8-OHdG简便检测方法,值得推广。 Objective:To observe results of detecting 8 - hydroxydeoxyguanosine (8 - OHdG) in urine by high - performance liquid chromatography. Methods: To detect 8 - OhdG level with the United States Agilent HPLC after dealing with urine. Results:The separation of 8 - OhdG's peak from impurities' was good, 8 - OhdG's concentration between 0. 5 μg/ml and 20 μg/ml was linear with the best mobile pH at 3.5, the recovery rate varied from 98.6% to 100. 1%. The difference (CV) was lower than 8%. Conclusion: HPLC is a highly sensitive, reproducible and easy method to promote detection method of the urine 8 - OHdG.
