摘要
目的:摸索高效液相方法测定10-羟基-α-癸烯酸的检测条件和方法,补充国家标准中检测方法的不足。方法:蜂王浆及其冻干粉中的10-羟基-α-癸烯酸经不同有机相提取后,采用不同流动相经C18色谱柱分离,紫外检测器检测。结果:采用无水乙醇+0.01 mol/L磷酸=5.5+4.5的10 ml混合液作为提取液,经超声处理,放置冰霜过夜沉淀取上清液试验,用甲醇+0.01 mol/L磷酸溶液=55+45作为流动相,检出限为1.6 mg/kg,在0~0.2 mg/ml范围内,线性关系良好,r=0.9996,RSD〈2%,加标回收率98%~101%。结论:该方法前处理简单、结果稳定、操作方便,符合大多数国家标准的定量习惯,可用于蜂王浆及蜂王浆冻干粉中的10-羟基-α-癸烯酸的测定。
Objective:To explore the detective condiction and approach of 10 -hydroxy-α-decanoic acid with HLPC to replenish the deficient in government standard. Methods: 10 - hydroxy - α - decanoic acid in royal jelly and lyophilized royal jelly are extracted with various organic phase, then isolated via C18 chromato collumn by diverse mobile phase, detected by UV detector. Results:The mixture of 5.5 ml dehydrated alcohol and 4. 5 ml 0.01 mol/L H3 PO4 were used for mobile phase uhrasounding. Overnight in refrig. Precipitate the supernatant were for detection. Use the methyl alcohol + 0. 01 mol/L phosphoric acids aqua =55 +45 as mobile phase. The detection limit was 1.6 mg/kg. The linear equation was good between 0 -0. 2 mg/ml,r = 0. 9996,RSD 〈 2%. The recover was 98 % - 101%. Conclusion: The pre - processing of the approach is simple, and consequence is stable and convenient to manipulate. Consistent with majority goverment standard hence it can be used to the detection of 10 - hydroxy - α - decanoic acid in royal jelly and lyophilized royal jelly.
