人参水溶性蛋白SDS-聚丙烯酰胺凝胶电泳指纹图谱研究 被引量:11

摘要 目的:建立人参药材的水溶性蛋白 SDS—PAGE 指纹图谱,为人参药材的质量评价提供依据。方法:采用 SDS-聚丙烯酰胺凝胶电泳法得到人参水溶性蛋白的电泳胶片,通过凝胶电泳图像分析软件生成谱图后,经计算机辅助相似度评价系统进行评价,并得到对照指纹图谱。结果:人参水溶性蛋白有9个特征带峰,12批次人参药材的相似度在0.90~1.0之间,稳定性、重复性和精密度良好。结论:该法具有实验设备简单、专属性强、灵敏度高、更直观的特点,可作为人参水溶性蛋白的 SDS—PAGE 凝胶指纹图谱,用于人参药材的质量评价。 Objective:To establish a fingerprint analysis and identification method for quality control of hydrophilic proteins from fresh Radix Ginseng which came from the different producing area and the different ages by constructing the SDS- PAGE -FP. Methods:SDS- PAGE was used with separate gel thickness 15% and concentrate gel thickness 5%. The standard proteins were reference. By similarity calculation after used to scan SDS - PAGE with Totallab, it was attained for the reference SDS - PAGE - FP of fresh Radix Ginseng. Results : There were nine peaks of Ginseng hydrophilic protein bands in the SDS - PAGE. The similarity of the superior and the ordinary in producing area were 0. 90 - 1.0 for the twelve kinds of fresh Radix Ginseng samples. The result was presented well with stability,repeatability and precision. Conclusion:The SDS - PAGE - FP of Ginseng hydrophilic proteins has been established and the feature of simple equipment, strong specificity, high sensitivity and direct - vision. The method can be used to identify and evaluate the quality of fresh Radix Ginseng from the different producing areas and the different ages.
