摘要
目的:寻找新的肝细胞癌相关基因。方法:采用差异显示技术对比研究肝细胞癌组织和与之相邻的非癌区肝组织信使核糖核酸的表达差异。结果:从2份肝细胞癌组织中获得46个表达差异的互补脱氧核糖核酸片段,克隆、测序分析了23个,对其中10个克隆进行了 RNA 酶保护法分析,仅 HCC3 9-22A 在肝细胞癌组织中高表达(5/8,62.5%)。结论:HCC3 9-22A 可能是一个候选的致癌基因,其进一步的意义正在论证中。
Objective:To identify new related genes in hepatocellular carcinoma.Methods:The mRNA expression of hepatocellular carcinoma was compared with that of adjacent nonneoplastic liver tissue by differential display technique.Results:Forty-six differentiaUy expressed cDNA fragments were present in two hepatocellular carcinoma samples,but absent in their adjacent liver tissues,and 23 of them were cloned and sequenced.Among 23 clones,10 were analysed on RNase protection,only HCC3 9-22A was confirmed to overexpression in 5 hepatocellular carcinoma samples(5/8,62.5%).Conclusion:HCC3 9-22A is more likely to be a candidate for oncogene.Its potential significance is under investigation.
