摘要
目的:研究牛磺酸对大鼠贮脂细胞、肝细胞增殖及前胶原基因表达的影响,为应用牛磺酸防治肝纤维化提供实验依据。方法:分离、培养大鼠贮脂细胞和肝细胞;采用~3H-胸腺嘧啶和~3H-脯氨酸掺入测定细胞增殖及胶原台成;用原位杂交检测Ⅰ、Ⅲ型前胶原 mRNA 含量。结果:在5~80mmol/L 浓度范围内,牛磺酸能显著抑制贮脂细胞、肝细胞的增殖及胶原合成,并呈时间、剂量依赖性关系;20mmol/L 牛磺酸可显著抑制Ⅰ、Ⅲ型前胶原 mRNA 的表达。结论:牛磺酸在体外具有抗肝纤维化作用,它对于防治肝纤维化可能有一定的临床意义和应用前景。
To study the effects of taurine on proliferation and procollagen gene expression of rat lipocytes and hepatocytes in vitro.Methods:Rat lipocytes and hepatocytes were isolated and cultured; cell proliferation and collagen synthesis were assessed by measuring the incoporation of ~3H-TdR and ~3H- proline;type Ⅰ and type Ⅲ procollagen mRNA levels were determined by in situ hybridization technique. Results:Turine (5~80mmol/L) markedly inhibited lipocyte,hepatocyte proliferation and collagen synthesis in a dose-and time dependent manner;20mmol/L taurine significantly suppressed the type Ⅰ and type Ⅲ procollagen mRNA expressions.Conclusions:Taurine has an antifibrotic action and could be significant and applicable for liver fibrosis.
