白色链霉菌JA3453中垩唑霉素生物合成基因簇调节基因的中断研究

摘要 通过生物信息学分析,在白色链霉菌JA3453菌株的垩唑霉素生物合成基因簇中定位了2个调节基因。其中1个是属于LysR家族的调控因子编码基因ozmR,另1个为编码转录激活因子基因ozmU。为了研究这2个调节基因的功能,选用垩唑霉素的产生菌链霉菌JA3453为实验材料,采用目标PCR的方法,对调节基因ozmR和ozmU分别进行基因中断。HPLC-MS分析显示,ozmR的中断可以提高垩唑霉素的产量,ozmU的中断则没有对垩唑霉素的产量带来明显影响。这些结论为利用调节基因提高垩唑霉素的产量提供了可能。 Oxazolomycin is a product from Streptomyces albus JA3453, In order to ascertain the response of regulatory gene on Oxazolomycin biosynthesis, disruptions of regulatory gene ozmR and ozmU were carried out separately. HPLC-MS analysis of the mutants revealed that disruption of ozmR would increase oxazolomycin's yield while disruption of ozm U has no obvious effect on oxazolomycin biosynthesis. The results suggest that ozmR is negative regulatory genes.
