摘要
目的:观察痛风胶囊对IgA肾病合并高尿酸血症血管内皮生长因子(VEGF)在肾组织表达的影响。方法:通过建立IgA肾病高尿酸血症的动物模型,同时给予痛风胶囊治疗,分别在第71、78、85、92、99天测24h尿蛋白定量,102d后检测血尿酸、血肌酐、血尿素氮、HE染色制片,光镜观察病理切片及采用免疫组化测定VEGF在肾组织的表达。结果:用药中、后期,治疗组、阳性对照组24h尿蛋白定量均低于模型组(P<0.05);用药后,治疗组、阳性对照组与空白对照组比较血尿酸无明显差异(P>0.05);阳性对照组血肌酐、血尿素氮平均水平明显高于各组(P<0.05);模型组和阳性对照组肾组织中VEGF表达强于空白对照组(P<0.05),治疗组的表达强度与空白对照组之间无显著性差异(P>0.05);阳性对照组肾小管坏死明显,治疗组肾组织结构基本正常。结论:痛风胶囊能降低血尿酸及尿蛋白水平,对肾功能及肾组织结构无损害,能减少VEGF的增生,改善肾血管的完整性。
Objective:studying Tongfeng Capsules intervening IgA nephrophathy with hyperuricaemia and the expression of vascular endothelial growth factor in nephridium, Method: through setting up the animal mould of IgA nephrophathy with hyperuricaemia, using Tongfeng Capsules for treatment, detecting the ration of 24 house' uric protein in 71,78,85,92 and 99 days respectively, after 102 days, detecting uric acid, nrea nitrogen, creatinine and using immunity to detect the expression of vascular endothelial growth factor in nephridium. Result: during metaphase and anaphase when using medicine, treatment and control of the ration of 24 house' uric protein are both under the model ( P 〈 0.05 ) ; After treating , compared with treatment and normal, uric acid is not obviously difference ( P 〉 0.05). compared with control and normal has the significance of statistics( P 〈 0.05 ) ; average of creatinine and nrea nitrogen in control is obviously higher than others(P 〈 0.05 ) ; the intensity of the expression of vascular endothelial growth factor in nephridium in model and control is higher than normal(P 〈 0.05 ), the intensity of treatment and normal has no obviously difference ( P 〉 0.05 ) ; morphometric analysis showed Allopurinol badly destroy nephron, however, Tongfeng capsules did not do that. Conclusion:Tongfeng Capsules can reduce the level of uric acid and uric protein: do no harm to nephridium; reduce the increasing of vascular endothelial growth factor and improve the integrality of nephridium vas.
