幼儿腭粘膜细胞钙离子浓度和龋病关系的研究 被引量:2

摘要 目的①测定无龋组、龋病低危组和龋病高危组幼儿腭粘膜细胞内钙离子的荧光强度;②分析细胞内钙离子荧光强度和患龋状况的关系。对象:50名4~5岁幼儿按患龋状况分为三组,无龋组22例(dft=0且CSI=0)、龋病低危组12例(0<dft<5且0<CSI<10)、龋病高危组16例(CSI≥10且dft≥5)。方法刮取受试幼儿腭粘膜表层脱落细胞,以钙离子荧光指示剂Fluo-3染色,用激光扫描共聚焦显微镜测定细胞内钙离子的荧光强度。结果①无龋组幼儿腭粘膜细胞钙离子荧光强度高于有龋组幼儿腭粘膜细胞内钙离子荧光强度,两组间差异有高度统计学意义(P<0.0001);②龋高危组幼儿腭粘膜细胞钙离子荧光强度均值明显低于龋低危组和无龋组,差异有高度统计学意义(P<0.0001);③不同性别间,钙离子荧光强度差异无统计学意义(P>0.05);④不同年龄间,钙离子荧光强度差异无统计学意义(P>0.05);⑤有龋组幼儿腭粘膜细胞内钙离子荧光强度与dft、dfs、CSI均无相关关系(rdft=-0.132,P>0.05;rdfs=-0.041,P>0.05;rCSI=-0.088,P>0.05)。结论4~5岁幼儿腭粘膜细胞内钙离子荧光强度与患龋状况之间有一定的关系。 Objective To explore the relationship between calcium ions concentration of palatine membrane cells and caries in children. Methods Fifty children aged 4 - 5 year - old ( boy 34, girl 16 ) were divided into three groups according to the caries status: caries free group (dft =0,CSI =0), low caries risk group (0 〈 dft 〈5, and 0 〈 CSI 〈 10) and high caries risk group (CSI≥10 and dft≥5). The palatine membrane cells were collected and died with Fluo -3. The fluorescence intensity of calcium ions in the cell was measured by laser scanning confocal microscope (LSCM). Results The fluorescence intensity of calcium ions in the caries free group was higher than that of the low and high caries risk groups with significant difference ( P 〈 0. 0001 ). The fluorescence intensity of calcium ions in the high caries risk group was lowest among the three groups ( P 〈 0.0001 ). No difference of calcium ions fluorescence intensity was found between gender and age ( P 〉 0.05 ). No correlation was found between the fluorescence intensity and dft, dfs and CSI indexes in the caries group, respectively (rdfh = -0.132, P 〉0.05;rdfs = -0.041, P 〉0.05;rCSI = -0.088, P 〉0.05). Conclusion The concentration of calcium ions in the palatine membrane cell is related with caries status in children.
