肝细胞肝癌组织TGF-β1和IGF-Ⅱ异常表达与HBV复制

Abnormal expression of TGF-B1 and IGF-Ⅱ expression associated with hepatitis B virus replication in hepatoma tissues

目的分析肝细胞肝癌(HCC)组织转化生长因子β1(TGF-β1)、胰岛素样生长因子Ⅱ(IGF-Ⅱ)表达与HBV复制间的关系。方法以自身对照法收集HCC组织及非癌组织,以生物素标记的HBV DNA探针检测HCC组织中HBV DNA,采用免疫组织化学法检测组织中TGF-β1和IGF-Ⅱ的表达,以巢式PCR扩增TGF-β1 mRNA和IGF-ⅡmRNA基因片段,并分析TGF-β1和IGF-Ⅱ表达与HBV复制间的临床病理学关系。结果HCC组TGF-β1和IGF-Ⅱ表达阳性率均为83.3%,TGF-β1 mRNA和IGF-Ⅱ mRNA阳性率均为100%,HCC组明显高于非癌组(P<0.01)。HCC组中TGF-β1和IGF-Ⅱ阳性表达与肿瘤分化程度显著相关(P<0.05),与肿瘤直径、数目间未见明显相关(P>0.05);TGF-β1和IGF-Ⅱ表达与HBV复制显著相关(94.7%),且HBV DNA阳性组显著高于HBV DNA阴性组(P<0.05)。结论HCC组织中TGF-β1和IGF-Ⅱ过度表达,且与HBV复制和肿瘤的分化程度有关。

Objective To investigate the relationship between hepatitis B virus （HBV） replication and the expressions of transforming growth factor （TGF）-β1 and insulin-like growth factor-Ⅱ （IGF-Ⅱ ） in hepatocellular carcinoma （HCC）. Methods Liver HBV DNA was detected by in situ molecular hybridization technique, and the expressions of TGF-β1 and IGF-Ⅱ were detected by immunohistochemistry, and TGF-β1 mRNA and IGF-Ⅱ mRNA were amplified by nested-PCR assay in HCCs and their self-control noncancerous tissues. The relationship was investigated between TGF-β1 or IGF-Ⅱ expression and HBV replication or their clinical pathological characteristics. Results The stronger expressions （83.3%） of TGF-β1 and IGF- Ⅱ were found, and the incidences of TGF-β1 mRNA and IGF- Ⅱ mRNA were 100% in HCC tissues. A significant difference was presented in the expressions between in HCC tissue and in non-cancerous liver tissues （P〈0. 01）. The positive rate of TGF-β1 in HCC was correlated to tumor differentiation, but neither to tumor size nor tumor diameter and numbers （P〉0. 05）. The levels of TGF-β1 and IGF-Ⅱ expression were significantly associated with HBV replication with higher in HBV-DNA-positive HCC （94. 7%） than that in HBV- DNA-negative group. Conclusion TGF-β1 and IGF- Ⅱ in HCC are overexpressed and associated with hepatic HBV replication and differentiation degree of HCC.
