碘普罗胺和碘比醇对大鼠骨髓间充质干细胞增殖的影响

摘要 目的:探讨碘普罗胺(IP)和碘比醇(IB)对大鼠骨髓间充质干细胞(MSCs)的影响。方法:将大鼠骨髓MSCs按2×10^3/孔接种于96孔板培养48 h后,改为IP或IB培养液(碘浓度分别为01、0、50、1002、005、001、0002、000μmol/L)各200μl继续培养,之后于不同时间点测定OD值;另外细胞按8×10^3/孔接种于24孔板,加IP或IB培养液各400μl孵育3 d后,计算细胞活力。结果:不同碘浓度IP培养液培养1 d、2 d、3 d和4 d不同时间点,各组OD值相比无统计学差异(P〉0.05),培养3d时,细胞活力在96.9%-98.2%之间;不同碘浓度IB培养液培养的不同时间点,各组OD值无统计学差异(P〉0.05),培养3d时,细胞活力在96.5%-98.1%之间。结论:IP和IB均不影响大鼠骨髓MSCs增殖,亦不增加细胞死亡。 Objective:To elucidate the influence of iopromide (IP) and iobitridol (IB) on mesenchymal stem cells( MSCs) proliferation of rat bone marrow in vitro.nethods:MSCs were cultured with IP or IB culture fluid, whose iodine density was 0, 10, 50, 100, 200, 500, 1 000, 2 000μmol/L, respectively, after inoculated in 96-well for 48h according to 2×10^3 cells per well, then OD value with MTT method were measured in different time point. Besides, MSCs were inoculated in 24-well according to 8 ×10^3 cells per well and cultured with above different density of IP or IB culture fluid for 3d, then cells viability were calculated with trypan blue repeling test method in different time point. Results: In the same time point, there aren't statistically significant( P 〉 0.05)about OD value of MSCs in different iodine density of IP culture fluid (cultured for 1 d, 2d, 3d and 4d, respectively), when cultured for 3 d, viability of MSCs was 96.9 % - 98.2 %. OD value in different iodine density of IB culture fluid, viability of MSCs was 96.5 % - 98.1%. Conclusion: Both IP and IB don't influence mesenchymal stem cells proliferation and increase their death of rat bone marrow in vitro.
