摘要
目的:研究糖尿病早期大鼠周围神经形态学变化,为糖尿病周围神经病变动物模型的确立提供客观依据。方法:采用链脲佐菌素(STZ)诱导大鼠建立糖尿病模型(DM),光镜及电镜下观察大鼠6周末坐骨神经病理形态学变化,并对坐骨神经横切面进行定量分析。结果:DM大鼠6周时坐骨神经出现明显的病理损害。光镜下见坐骨神经有髓鞘神经纤维空泡样变性、轴索肿胀。电镜下见严重脱鞘改变。病理图像分析结果与电镜结果相符。结论:STZ诱导大鼠早期已出现明显的周围神经损害,符合糖尿病周围神经病变(DPN)的病理特点,说明DPN模型建立成功。
Objective: To study the peripheral nerve changes in diabetic rats in the early stage, in order to proffer an objective and theoretic reference for the establishment of animal model being diabetic peripheral neurophathy. Methods: The models of rats were induced by Strepotozotocin (STZ). After 6 weeks, the histomorphometry of sural nerve was analysed with light microscope and electron microscpe. Results: Morphometric analysis showed that the fibers density in the nerve of diabetic rat was significantly fewer than that of normal group ( P 〈 0. 01 ). Conclusion: Peripheral neurophathy of STZ-induced diabetic rats appeared in the early stage, corresponded with the histomorphometry of diabetic peripheral neurophathy. It certifies that the model of diabetic peripheral neurophathy is successful.
