摘要
目的探讨葛根素对大鼠脑缺血后脑促红细胞生成素(EPO)表达的影响。方法应用线栓法制备大鼠大脑中动脉缺血2h再灌注48h损伤模型;分别于缺血2h再灌注即刻、再灌注24h给予腹腔注射葛根素400mg/kg(体重),对照组腹腔注射等剂量生理盐水,每组12只大鼠。大鼠苏醒后30min及处死前分别进行神经功能评分。48h处死大鼠,每组6只石蜡切片用HE染色检测腔梗死体积,免疫组织化学检测葛根素对缺血脑组织EPO表达的影响;每组6只ELISA法检测EPO活性。结果葛根素400mg/kg(体重)腹腔注射组的脑梗死体积明显小于对照组,葛根素治疗也可减少神经功能损伤,神经功能评分均明显好于对照组。免疫组织化学结果显示:葛根素组大鼠脑缺血侧皮层区及基底区EPO阳性细胞数明显高于对照组。结论葛根素对大鼠脑缺血再灌注损伤具有保护作用。其保护作用可能与提高EPO的表达有关。
Objective To test whether Chinese medicine puerarin has effects in erythropoietin (EPO) expression in rats following middle cerebral artery occlusion (MCAO). Methods Ischemia-reperfusion injury ( I/R injury) was induced by ligated MCAO with 2 h ischemia and 48 h reperfusion. 400 mg/kg puerarin or saline (control) were administered intraperitoneally at two points: immediately and 48 h after reperfusion. The rats were sacrificed 48 h after reperfusion. Infarct volume was measured through triphenyl tetrazolium chloride (TTC) staining. The EPO expressions were determined by immunohistochemistry. Functional neurological outcome was determined at30 min and 48 h after the rats awake from the surgery. Results Compared to control group, the infarct volume of 400 mg/kg puerarin group decreased obviously. The puerarin-treated group had better functional neurological outcome compared to the controls. The EPO expressions were increased obviously in puerarin-treated group. Conclusions Puerarin has neuroprotective effects on rats with I/R injury, one of the underlying mechanism may through increasing EPO expressions during I/R injury.
